A rapid point-of-treatment diagnostic assay for HIV-resistance to 1st-line ART

The primary goals of the project are to:

(1) Concentrate the HIV DNA from blood specimens via stimuli-responsive reagents to enable detection of drug-resistant mutants comprising ≥10% of the HIV population

(2) amplify HIV DNA using isothermal strand displacement amplification to levels sufficient for detection of drug-resistant mutants comprising ≥10% of the HIV population

(3) develop a one-pot ligation that targets HIV codons relevant to 1st-line ART through analysis of probe competition and synthesis of modified ligation probes

(4) develop a simple and rapid paper strip test for multiplexed detection of all codons at 10% mutant levels

(5) combine re-engineered protocols into a rapid and simplified kit (OLA-Simple)
